From the pylote site:

on the other hand, the menu File → Create Launcher will allow you to create a file pylote.desktop wherever you want (desktop, menu …).

Create that pylote.desktop file in /usr/share/applications and it should automatically show in jgmenu. Although, I don't know what section it will appear in.

Do what PackRat said.
You can put the desktop file in $HOME/.local/share/applications too.
Or edit $HOME/.config/jgmenu/prepend.csv and just add pylote as the first line

Thanks, I will try to put the file and see if work or I will try to edit the csv on first line as both recommendations.
